Combined influence of sulfur dioxide and allergenic products of microbiological synthesis of mycotic genus candida on neutrophils of guinea-pigs (sulfur dioxide, sensibilisation and neutrophil metabolism)

Purpose: To study the functional-metabolic features of the blood neutrophiles in the guinea-pigs undergoing to the isolated and the combined exposure to the sensibilizing factor (proteinconteining dust – PCD) and sulfur dioxide (SO2).
Methods: The neutrophile percent capable to uptake the latex particles as well as average number of the latex particles captured by the phagocytar neutrophiles (phagocytar index – PhI) were determined. The intracellular levels of the alkaline phosphatase (AlP), acid phosphatase (AcP) and the glycogen content were parallel taken into account and the results were expressed as the conventional units according to L. Kaplow. The sensibilisation state was assessed using the responsiveness of the passive cutaneous anaphylaxis and the intracutaneous allergic samples with the polysaccharide antigen, isolated from the product of the microbiological synthesis yeast-like fungi of the genus Candida.
Results: The differentiated changes have been revealed in the blood neutrophiles in dependence on the exposure specific character and on the expression degree of the hypersensitivity (HS) responses. The values of the intracellular parameters in the group of the intact animals were found to amount on the average 1,91±0,05 (AlP), 2,06±0,02 (glycogen), 0,35±0,04 (AcP) and 5,66±0,40 (PhI). Isolated administration of the PCD was observed to lead to the decrease and the isolated administration of SO2 – to the increase in the levels of AlP (up to 1,68±0,05 and 2,42±0,08) and glycogen (up to 1,89±0,03 and 2,47±0,06). In the combined exposure to PCD and SO2 the indices above may response only in the case of absence of the HS stimulation, increasing in (AlP) and reducing in (glycogen) and were without changes compared with the control in increasing in the allergotest levels. On the contrary, the activation of AcP was noted to express in the larger degree after the combined administration of PCD and SO2 followed by the decrease in PhI. Thereby, under condition of progressing the HS the AcP level was maximal (1,16±0,03), the level of PhI – minimal (2,94±0,17). The participation of the compensatory mechanisms is suggested to be in the modification of the metabolic neutrophile status in association with the increase in the sensibilization signs to the biological allergen. |
